- Therefore, this is the third season to feature all-male teams in the final three (tied with The Amazing Race 3 (Latin America) and The Amazing Race 4 (Latin America)). This would later be followed by The Amazing Race 35.
- First time that a team used the U-Turn twice.

- First time visits to Chile, Argentina, and India.
- First (and only) season to visit the continent of South America.
- First season to feature a transgender contestant (Hamilton Elliott).
- First season to feature non-consecutive Pit Stops in the same province.
- First season to feature a Blind U-Turn where one team can remain anonymously (without putting up their own team sticker) and may force another team to complete the other option of the Detour that was not originally picked.
- First introduction of the Face Off in which two teams must compete against each other in a challenge.
- The team that wins receives their next clue, while the team that loses must wait for another opponent and start over. Ultimately, the last team remaining at the challenge must serve a pre-determined time penalty before they can continue racing.
- First season where all-male teams survived from start until the end.
